장비소개

900 MHz 핵자기공명분광기 장치는 생체분자 입체구조 규명 및 신약개발연구의 핵심 장비 로 활용되며 이를 오창센터에 설치하여 국가적 공동 활용 연구 장비로 활용하고 있습니다.

장비특성

  • 초저온 프로브의 1H 민감도는 기존 실온 프로브의 4배 이상(8,000)으로 실험시간을 1/16으로 단축
  • 100 μM 이하 단백질 측정 가능
  • 100 μg 천연물의 13C 실험 가능

대표연구성과

세포막 결합된 TRPV1 채널의 독거미 독에 의한 활성 메커니즘 규명

TRPV1은 double-knot toxin(독거미 독)이라 불리는 단백질에 의해 활성화 됨. 채널 활성화 메커니즘 규명을 위해 독거미 독소의 3차 구조를 밝힐 필요가 있음. 좋은 해상도의 NMR 스펙트럼을 위해 900 MHz NMR을 활용하여 2D-NOESY 등 2차원실험을 수행하여 정확한 3차 구조를 규명함
